DNA FISH validation of Sat11 (in green) and telomere (in red) elimination in embryos. DNA is in blue; scale bars, 10 and 2 m for embryos and insets, respectively. The probes are detected in all nuclei of a 2-cell embryo, including the polar body (upper embryo). In a 7-cell stage embryo (middle embryo), the probes are visible only in the C and P3 blastomeres. In metaphase of a division of elimination (here EMS), the probes are found only in fragments outside the metaphase plate. Similarly, after a division of elimination (daughters of ABa and ABp) the probes are found only in the DNA fragments outside the nuclei. In an ~32-cell stage embryo (bottom embryo), the probes are visible in a single nucleus, most likely P4. The white stars show the polar body. A single focal plane is shown except for the top embryo, which shows a Z projection of all focal planes. See alsoFigures S2andS3.
