Immunoblotting analysis of Wis1 and Sty1 phosphorylation in cells expressing wild-type Sty1 (MC2) or Sty1-Tpx1 fusion protein (MC12) before and following 10 min exposure to increasing concentrations of H2O2. The lower mobility of phosphorylated Wis1 was detected by immunoblotting proteins separated using Phos-tag SDS-PAGE. The relative levels of dual TGY motif phosphorylated Sty1 (anti-ppP38)/total Sty1 (anti-Hog1) normalized to wild type (t = 0) are shown below each lane.
