Schematic illustration of genetically encoded cis -active SAMURI. HEK293T cells grown in a cell culture dish were transfected with plasmid, and the cSAMURI-expressing cells were treated with ProSeDMA. The intracellularly propargylated RNA was isolated and labelled with Cy5 azide. The modification yield was quantified based on the fluorescence intensities of Cy5 and DFHBI-1T.
