Schematic of virus-endosome fusion assay based on fluorescence self-quenching. Fluorescence associated with R18 probe-labeled virus particles is quenched due to dye-dye interactions. Upon virus entry, virus membrane fusion mediated by a drop in endosomal pH causes dispersion of the probe, resulting in increased fluorescence intensity due to de-quenching mediated by monomerization of the probe.
