Workflow from Scientific Research

Overview of PE systems. All use a prime editor protein consisting of SpCas9(H840A) nickase fused to a reverse transcriptase (RT) enzyme. PE1 uses the wild-type RT from the Moloney murine leukemia virus (M-MLV), while the PE2 system uses an engineered pentamutant variant of the M-MLV RT. PE3 uses an additional single guide RNA (sgRNA) to nick the non-edited strand. PBS = primer binding site. RT template = reverse transcriptase template.
