Orphan quality control establishes network dynamics required for gene expression. Transcription factors (TFs) in the c-Myc network are continuously produced, form complexes with stabilizing partners (SPs), and drive gene expression. Following dissociation, UBR5 degrades one subunit to render complex disassembly irreversible. The stabilizing partner can be re-used, whereas the unstable subunit must be re-synthesized. This network architecture allows transcription factors to efficiently switch from early to late partners while remaining susceptible to stress-induced inhibition of gene expression.
