Left: an example set of RRM domains. We choose domains that are expected to bind single-stranded RNA in a canonical binding mode, as they contain three hallmark conserved hydrophobic residues (black arrows and right part of this panel). The green arrow indicates the position where a Lysine or an Arginine is found in high RNA-binding affinity RRM domains. We propose that, in the domains whose seqeunces are displayed, this residue could be mutated to Lys or Arg in order to obtain an affinity enhancement. Right: canonical RRM-RNA binding mode, the relevant contacts are represented.
