Isolation of CLDs from Nicotiana leaves agro-infiltrated with the EV (control) or a construct encoding tHMGR, AL-1, AL-2, and AL-3 (TA123 combination) using sucrose density gradient centrifugation. The less dense CLDs float from the extract phase and through two floating buffers to accumulate as a milky band visible on top of the liquid. TA123 CLDs display an orange-red color, also observed by light microscopy, while those from EV leaves are almost colorless. Nile red staining of CLDs was shown in Supplemental Figure 6A.
