Calculation of the second derivative of AP waveforms reveals that the axon initial segment (AIS) spike is largest in amplitude at an axon-bearing dendritic recording site (blue, upper traces) and attenuates as it spreads to the soma (black, upper traces), preceding the somato-dendritic component. Inversely, in a recording made from a non-axon-bearing dendrite the AIS spike is largest in amplitude at the soma (lower traces). Same neurons as illustrated in (A).
