Identifying the relevant population (yellow shaded region), key goals of the study (green shaded region; variant detection or measuring variant prevalence), and sampling method (blue shaded region; cross-sectional or periodic) are necessary to determine the required targets and parameters (red shaded region) that must be specified in order to calculate the appropriate sample size. Importantly, parameter estimates should be specific to the pre-defined population of interest as well as reflective of the logistical and biological sources of bias at the time of sampling.
