A speculative model for improving the editing efficiency of mtDNA by combining nickase with deaminase (take TadA8e-V106W as an example in this model). TALE–nickase binds the target DNA and nicks the dsDNA. The nicked dsDNA may be prone to form ssDNA structures. TALE–TadA8e-V106W binds the target DNA and efficiently deaminates the adenine(s) on the resulting ssDNA. The resulting inosine(s) can be converted permanently to guanine(s) after DNA repair or DNA replication.
