Normalized binding of MRCK1955-1534-EGFP (C1-PH-CNH) to liposomes containing the indicated lipid species in a high-throughput LiMA assay. Binding was quantified as the ratio of MRCK1955-1534-EGFP to Atto647-conjugated phosphatidylethanolamine incorporated into the liposomes. MRCK1 binding to each lipid was evaluated with three different concentrations of signaling lipid, yellow (2 mol %), orange (5 mol %), and red (10 mol %), and compared with the PH domain of PLC, which binds specifically to PI(4,5)P2and was used as a positive control, cyan (2 mol %), blue (5 mol %), and purple (10 mol %).
