In vitro fatty acylation of recombinant TEAD1 proteins. WT, KR, or CS mutant recombinant His-tagged TEAD1-YBD (YAP-binding domain) proteins purified from E. coli were incubated with 1 uM alkyne-palmitoyl-CoA (Pal-CoA alkyne) or palmitoyl-CoA (Pal-CoA) as a control, followed by click chemistry with FLAG-azide. Fatty-acylated TEADs were detected by western blotting with an anti-FLAG antibody.
