lentiMPRA strategy for large-scale libraries. Thousands of cCREs including potential enhancers (marked by DNase peaks) and promoters (centred at the transcription start site (TSS)) are inserted into a reporter plasmid in both orientations together with barcodes. The libraries are infected into cells using lentivirus and the integrated DNA and transcribed RNA barcodes are sequenced to quantify cCRE activity.
