Schematic illustration of the different steps of the NanoB2approach. After genetic fusion of the membrane protein of interest to NanoLuciferase (1), a nanobody against this target protein is obtained from public online databases or screening campaigns (2). Subsequently, nanobodies are labeled with a fluorescent molecule of choice (3) and equilibrium binding of nanobodies can be tested using live whole cells or membranes expressing NanoLuc-tagged receptor in a homogeneous NanoBRET readout (4).
