Schematic of experimental outline. Primary cortical astrocytes were isolated from postnatal P3GFAP-CreERT2; LSL-tdTomato; p53flox/flox(p53Gfap-icKO) pups. Following immunopanning, astrocytes were incubated with 4-hydroxytamoxifen (4OHT) in EGF/FGF media for 5 days to inducep53and tdTomato allele recombination and astrocyte dedifferentiation, before collection for 10x Genomics scRNA-seq.
