Median Brillouin shift averaged volumetrically over three distinct tissue regions (epidermis, central nervous system (CNS) and endoderm–mesoderm) in n = 3 embryos, manually segmented in 3D according to SPIM data. Embryo 3 is shown in g. There was no significant difference in Brillouin shift between the segmented embryonic domains (P = 0.1944, one-way ANOVA, paired, non-parametric test (Friedman test)). Each dot represents the median, and the lines represent the interquartile range of segmented domains in each analyzed embryo (n = 3 embryos from independent experiments). The green, yellow and light-blue outlines in g and h show the segmented regions for epidermis, CNS and endoderm–mesoderm, respectively.
