Schematic of the mouse model for gene editing on therapeutic T cells in vivo. C57BL/6J mice were injected subcutaneously with 2 105 OVA-expressing B16 cells followed by an intravenous injection of 5 106 Cas9/OT-1 T cells at day 12 after tumor injection. At 15 days after tumor injection, mice were injected intravenously with 1 1012vg/mouse Ark313 expressing either an NTC sgRNA or Zc3h12a-targeting sgRNA, and an mCherry reporter gene.
