Schematic of the hepatic PC-catabolic pathway. PNPLA8 and PNPLA7 hydrolyze PC and LPC, respectively, cooperatively producing GPC. This pathway is required for mobilization of endogenous Cho from membrane PC, allowing flux of methyl groups into the Met cycle, and is also linked to PC re-biosynthesis (PC recycling), polyamine synthesis, transsulfuration, and histone/DNA methylation as well as TG synthesis via G3P. Pnpla8-/- and Pnpla7-/- mice show similar phenotypes reminiscent of Met insufficiency. Mean SEM;*p < 0.05,**p < 0.01, and***p < 0.001 versus Pnpla8+/+ mice.
