Mutations that may interfere with the conformation of the N-terminal cap impaired GPR20’s basal activity. ΔBRET: the change of bioluminescence resonance energy transfer value (Materials and methods). The ΔBRET was compared between the wild-type (WT) GPR20 and various mutants. Significance was determined by two-way analysis of variance (ANOVA) without repeated measures, followed by Dunnett’s post hoc test (*** P < 0.001, ** P < 0.01). Data are mean ± s.e.m.
