Schematic workflow of spatial ATAC. Transposition with Tn5 is performed on immunostained tissue cryosections immobilized on a barcoded slide (55 m spot diameter; 100 m interspot distance). Transposed fragments are surface-captured using a splint oligonucleotide, which is ligated and extended to allow the generation of a spatially barcoded DNA library.
