Representative flow cytometry histograms of cells expressing the TEV substrate (TEV-control), treated with either matriptase or TEV protease. Cells stained with the cleavage reporter but not treated with protease served as a positive control (denoted: no protease), while cells treated neither with cleavage reporter (no HER2-biotin) nor protease served as a negative control (denoted: cells). The experiment was performed in triplicate (n= 3), and corresponding bar plots show the mean SD. TEV-treated cells exhibited significantly lower signals from the cleavage reporter compared to both untreated cells (p= 0.009) and Mtp-treated cells (p= 0.009). No significant difference was observed between Mtp-treated and untreated cells (p= 0.47).
