For recordings without STIM1, currents were taken before and after applying 15 s of UV light (top). In the presence of STIM1, UV light was applied after complete STIM1-Orai1-UAA binding (bottom). For experiments with STIM1 (in D, E, G, and H), time points of maximum currents after [Ca2+]ER↓ store depletion and after the additional application of UV ([Ca2+]ER↓+ UV) were taken. UV light was applied for 15 s after maximal store-operated currents were reached.
