Non-treated HCV-SGR cell. Cells bearing a subgenomic HCV-SGRmCherryreplicon and parental Huh7 cells were seeded onto Au quantifoil grids, labeled with MTG, and cryofixed by rapid vitrification in liquid nitrogen. Grids were subsequently processed first for cryo-fluorescence microscopy and for cryo-SXT in the SXT-100 table-top instrument to enable correlative imaging FL-SXT. Left panels represent cryo-epifluorescence microscopy imaging (epifluorescence). NS5AmCherrysignal is shown in red and MTG is shown in green. Right panels show a volume slice of tomogram corresponding to the same region (SXT volume slice). Overlap of the two signals enables identification of viral and cellular structures (central panels; overlay). White arrows indicate the presence of high contrast pleomorphic NS5A-positive structures. Black arrows indicate low contrast mitotracker-positive structures. Scale bars, 5 m. SeeVideo S2.
