Secretions obtained from mice in each of the two groups (A–C) were pooled, divided into two equal aliquots that were treated with either neuraminidase (specific for 2-3-linked sialic acid) or neuraminidase buffer, and then captured on microtiter wells coated with polyclonal rabbit anti-N. gonorrhoeaeH.8 lipoprotein antibody. Detection of LOS sialic acid on capturedN. gonorrhoeaeusingMaackia amurensisII (MAL II) lectin. H.8 captured bacteria were incubated with biotinylated MAL II lectin, and binding was visualized using avidin alkaline phosphatase. Control neuraminidase-treated bacteria did not bind MAL II.
