A single RNA Pol II stalled elongation complex, ligated upstream of a molecular ruler and nucleosome, is tethered to two beads held in optical traps. Transcription is restarted by the addition of nucleotides in the presence or absence of FACT and is monitored in real time as tether extension changes under constant force.
