Pitstops impair CCP dynamics in the perinuclear region. After photobleaching of eGFP-clathrin-LCa in a circular region of interest fluorescence recovery was determined over time (seeFigure S2C for exemplary images). Fluorescence intensities were normalized to initial values obtained before bleaching, and plotted over time (seeFigure S2D). Maximal recovery was determined by hyperbolic curve fitting (DMSO: 75.5%; Pitstop 2: 54.4%; Pitstop 2c: 44.4%; Pitstop 2d: 37.6%). Values are represented as mean ± s.e.m. from at least 15 cells.
