Model summarizing current understanding of signaling and trafficking pathways of LHR activated by LH agonist. Upon activation, LHR couples to interactors that mediate its downstream effects including cAMP signaling, desensitization and internalization. Following internalization, LHR is trafficked to very early endosomes (VEEs), smaller in size than early endosomes and lacking classical endosomal markers EEA1 and Rab5. From the VEE, LHR elicits downstream signaling cascades, including cAMP generation. LHR is then primarily rapidly recycled back intact to the plasma membrane, with a minor proportion sorted toward the early endosome and subsequently degradation.11However, the dynamic LHR interactome during endocytic trafficking to and from VEEs is currently unknown.
