Validation of LHR-APEX2 signaling by TR-FRET cAMP assay normalized to the LHR WT Emax. Dose-response cAMP curves for the FLAG-LHR WT HEK293 cells and tetracycline-induced Flp-In T-REx 293 LHR-APEX2 cells activated by various concentrations of LH produce comparable EC50and Emaxvalues. Non-induced cells do not respond to LH treatment. Data represent mean ± SEM (n= 3).
