Phosphomimetic (glutamate; E) but not serine- or threonine-to-alanine (A) mutations of the 23 TORC1 sites in Tco89 cause mild rapamycin sensitivity. Wild-type (WT), tco89Δ, tco8923A, and tco8923E cells were grown exponentially, spotted (10-fold serial dilutions) on SD-LHUAW plates without (control) or with rapamycin (+Rap; concentrations indicated), and grown for 3 days at 30°C.
