Epithelial cell monolayers infected with wild-type parasites transfected with plasmids driving altered MVP1-HA. Cell monolayers were fixed and labeled at 24 h post-infection. Note that removal of the predicted signal peptide and mutation of the conserved region LSFV both caused a block in host export. Further point mutations demonstrated that mutation of phenylalanine alone, F39A, was sufficient to disrupt trafficking into the infected host cell.
