Serial dilution spottings on yeast extract pentose dextrose (YPD) media of wild-type (WT) (-) or anchor-away strains. WT or stm1anchor-away strains carry an extra copy of genomically integrated RPO21 WT (RPO21) or different point mutations (rpo21N479S, rpo21N1082S, rpo21N1085Q). Nuclear depletion of endogenous RPO21 (anchor-away) was initiated by the addition of solvent control (- anchor-away) or 1 M rapamycin (+ anchor-away), and cells were incubated (3 days, 30C).
