In vivo pulse-chase proximity biotin ribosome labeling system. During the pulse phase (on), the 60S subunit (blue) protein Rpl16a/b-hemagglutinin (HA)-Tobacco Etch virus protease site (TEV)-peptide tag for enzymatic biotinylation (AVI) is biotinylated (purple star) by the estradiol-inducible BirA ligase. BirA degradation by the auxin-inducible degron abolishes further biotin labeling (off). Biotinylated 60S ribosomes are chased (CHASE) over time and detected by streptavidin.
