Uracil in DNA can be a consequence of spontaneous deamination, generating a mutagenic U:G mispair, or uracil can be misincorporated instead of thymidine; a consequence of an altered dTTP/dUTP pool balance. Alternatively, uracil in DNA is enzymatically generated through the deamination of cytosine on unprotected single-stranded DNA (ssDNA), primarily by APOBEC3A or APOBEC3B.
