Sucralose dose responses of TAS1R2 mutants (Y756A and F827A) in cell-based calcium imaging assays. Cells were co-transfected with WT or mutant TAS1R2 together with TAS1R3 and Ga15/gust44 (seeSTAR Methods). Data are mean ± SEM ofn= 6–8 independent replicates. The WT data are as shown for the sucralose responses inFigure 2E. The Y756A mutation abolishes receptor activity. The F827A mutation reduces maximal response but has no effect on EC50, as would be expected from a mutation that impairs receptor-G protein binding but not agonist affinity. See alsoFigures S2andS8.
