Percentage of insulin mRNA left over after 1 h in vitro cleavage with recombinant IRE1α for donors with INSS/S (red, three donors), INSP/S (blue, three donors), or INSP/P (light blue, one donor). For statistical analysis, data from the single and rare homozygous INSP/P islet donor were combined with those from heterozygous donors. Delta Ct values were calculated by subtracting the mean Ct values of RNA samples treated without IRE1α from samples treated with IRE1α. Percent relative expression was calculated by 2−dCt × 100.
