REV7-REV3-REV1 complexes are recruited to some of the lesions where replisome progression has been blocked. In these complexes, REV7 adopts a closed-state conformation that catalyzes the conversion of free (apo) REV7 molecules from the open (gray) to the closed (yellow) state. In this process, the converted REV7 molecules capture a protein (green) that regulates entry into mitosis. As a result, entry into mitosis is inhibited.
