RAW cells were labeled for 2 min at 37°C with DHE/MBCD complex, washed, and chased for 8 h in culture medium (with LDL-depleted serum) in the presence of 7-KC (10 uM) or ethanol (EtOH). Non-polarized RAW M0, and M1 or M2 polarized cells (the same protocol of polarization as for BMDMs,Figure 2) were first labeled with DHE (in green). Wheat germ agglutinin (WGA) was added after the chase to visualize the plasma membrane (in red). The number of macrophages analyzed was for M0: EtOH (n= 25), 7-KC (n= 37); M1: EtOH (n= 40), 7-KC (n= 31); M2: EtOH (n= 37), 7-KC (n= 39). Data are represented as means +- SEM. A Mann-Whitney U test was used for statistical analysis with****p< 0.0001.
