Imaging neurotransmitter release with a pHluorin probe. Synaptophysin-pHluorin (SypHy) is expressed on the luminal face of synaptic vesicles, where its fluorescence is quenched at intravesicular pH 5.5. Exocytosis exposes the luminal face of the vesicle to extracellular pH 7.4, unquenching SypHy fluorescence (green).
