Schematic of differentiation of dKOEoGFP cells in the presence or absence of ME-inducing signals using 3 conditions: (1) ME induction by activin A (ActA, 30 ng/mL) in absence (nodox), (2) in presence (+dox, 5 μg/mL) of EOMES:GFP, and (3) in presence (+dox) of EOMES:GFP while blocking NODAL/TGF-β-signaling by SB431542 (SB, 10 μM) and Wnt-signaling by XAV939 (XAV, 2 μM). Cells were harvested at day 3 after 24 h of treatment for RNA-seq, ATAC-seq, and H3K27ac ChIP-seq.
