MMC sensitivity was assessed on samples from (E), except SAP-2. On the left side, domains that did not rescue FANCA protein expression and splicing were analyzed with increasing doses of MMC. The black constant line (sgNT wild-type control) and the blue constant line (full-length CCAR1-/- complement) indicate the positive controls, while the pink constant line (knockout, KO) represents the negative control. On the right side, domains dispensable for FANCA protein expression and splicing were analyzed with increasing doses of MMC. The same controls were used in both plots. The graphs summarize the mean of 3 biological replicates and error bars indicate standard deviation.
