Analysis of Nth1 PKA-dependent phosphorylation in WT andtpk1 tpk2cells. Soluble Nth1 phosphorylation was detected using a phospho-PKA (RRXS*/T*)-specific antibody. Total Nth1 protein was probed using a FLAG antibody. Quantification is shown as the ratio of phospho-Nth1 to total immunoprecipitated Nth1 (P-Nth1/Nth1). Data shown are the means of at least three independent biological replicates SD; WT values are based on seven biological repeats across experiments shown here and inFigures 6G and 6H to provide a baseline across experiments. Significance is indicated relative to the respective untreated WT ortpk1 tpk2control (*p< 0.01,***p< 0.001; ns, not significant; one-way ANOVA).
