Diagram of the experiment. Monolayers were treated with calyculin, an inhibitor of myosin phosphatase, at t = 0. The tension in the monolayer was measured over time and the length of the monolayer was kept constant by the micromanipulator. After some time, defects appeared in the monolayer and measurements were continued until the monolayer failed. The dashed black line indicates a representative position at which the monolayer profile would be imaged.
