Ribosome stalling by rumicidins on ermCL mRNA, as revealed by the reverse-transcription inhibition (toe-printing) assay in a cell-free translation system. Thiostrepton (THS) and Bac7[1-22] were used as control compounds inhibiting the elongation step of bacterial translation. Both of them allow ribosomes to initiate translation at the AUG start codon but prevent further progression of the ribosome either due to the blocked access of elongation factors by THS 76 or due to the inability of aminoacyl-tRNA to accommodate into the A site by Bac7 5 . Two independent experiments were performed, and the observed effects were the same. Source data are provided as a Source Data file.
