The component with sTb(C)-M13-GFP is tethered to the membrane via the transmembrane domain of a CD4 cell membrane protein 58, while the CaM-V5 epitope tag 59-sTb(N) component is expressed throughout the cytosol. CaST only tags proteins when cells are treated with biotin and exhibit elevated intracellular Ca 2+.
