Using Cy3B as a reference for each cell and dye, the relative specificity (that is, whether a dye exhibits less or more off-target localizations than Cy3B) is plotted for all examined dyes. Except for CF640R, Atto655 and Janelia Fluor 585, all dyes exhibit increased off-target binding compared with Cy3B. The circles represent the mean relative specificity, and the error bars represent the s.d. across 3 repeats per dye.
