The C-terminal tails of p97 were fused to HaloTag domains and linked to cPA FAM strands. The hexameric p97cPA FAM conjugate (780 kDa) was encapsulated into the NE compartment via hybridization to six PA strands extending from the inner surface of the origami, leaving 6-nt-long toeholds to facilitate protein movement and permit subsequent protein displacement.
