Yields achieved using circular unstructured LEAPER guide RNAs containing a 111-nt-long antisense part. The basic design (column circular LEAPER) does not contain bystander solutions. The other guide RNAs either contain GA mismatches or apply U depletion at all bystander sites, GU wobbles at GU-amenable sites or a combination of GA mismatching or U depletion with GU wobbles at amenable sites.
