Naive cells from Tcf7-YFP mice were stimulated with alphaCD3 and alphaCD28 (TCR stim) for 2 days with IL-2, IL-7, IL-15, and IL-12. At day 2, TCR stimulation was removed but the same cytokine cocktail was used. At day 3, cells were sorted for Tcf7-YFPlo and Tcf7-YFPhi populations and recultured ex vivo, with and without continued TCR stimulation and IL-12 in the presence of IL-2, IL-7, and IL-15. Light and dark blue coloring throughout correspond to sorted Tcf7-YFPlo and Tcf7-YFPhi populations, respectively.
