Workflow of the optical tweezer force clamp and force-extension experiments. Force clamp experiment: the force applied to the tethered DNA was kept at a constant value (1 pN) as PARP1-dsDNA co-condensation would result in the shortening of the tether. The DNA used was nicked on both strands by CAS9(D10A) with a 60-bp gap between the nicks. Force-extension experiment: PARP1 condensates were formed on tethered DNA and the tether was pulled apart at a constant speed (0.1 m/s) until rupture occurred.
